Ihr Broadway-Debüt 2003 in einer Wiederaufnahme des Musicals „Chicago“ wurde nach ihrer erfolgreichen Theaterkarriere in London mit großem Beifall aufgenommen.
Griffith kehrte in den 2010er Jahren auf die Leinwand zurück und trat in zwei Filmen auf: dem Science-Fiction-Film Autómata (2014), in dem sie zusammen mit ihrem Ehemann Antonio Banderas die Hauptrolle spielte, und James Francos The Disaster Artist, in dem sie eine Rolle spielte Ausbilder (2017).
