Barbara Mori nació en una familia acomodada. Su padre, Yuyi Mori, es un empresario inmobiliario. Su madre Rosario Ochoa es empresaria. Su hermana Kenya Mori es modelo y actriz. Su hermano Kintaro Mori es actor y modelo. Anteriormente, salió con el actor y político Sergio Mayer y tiene un hijo llamado Sergio, pero la pareja nunca se casó.
También tuvo romances con el actor Manolo Cardona, el beisbolista Kenneth Ray y actualmente tiene una relación con el diseñador María José Torre.
